727 trany problem I just installed it

I just put a used 727 in my j-10 it has 150,000 miles on it it sat on the ground for about 2 weeks I pulled it out and put it back in my J-10 it now sits behind a 401 it used to sit behind a 360 my problem is I have problems with it shifting in to any gear at all. I have to take a line off the radiator and wait for it to pump some fluid out and once it starts I put it back on and I can drive it for ever but then when I park it and let it sit it wont work the next time I get in it. any suggestiond would be appreciated I just changed the filter to and it has the appropriate fluid level it also has a huge axuilary cooler. should I take it to get flushed or what?

Need a little more information, I'm assuming that you mean that you can physically engage the shift selector into D or R, but the vehicle doesn't move. right?
