in short it's the distance from the rear lip of the rim to the mounting surface of the rim. If you google backspace you'll get a ton of inof and how-tos.

The wheel mounting surface (back side of the rim where the bolt holes are) is five inches from the inside edge of the rim. It is a measure of where the tire sits compared to where the wheel is mounted. On a low backspacing wheel, the tires will sit further out from the vehicle.
