That Indian 455 would be pretty stinkin' reliable. I don't think you'd have much trouble finding parts, but maybe? They are getting kinda old with the last one built in 1975.
I had a friend with a '69 GS 400, 3.08 rear and a Muncie knock down about 20 mpg pretty regularly. The 400 is the same design as the Buick 430-455. With some mild gears (say 3.55s?) and the 6 speed, I'd be you could pull some mileage out of a big cube motor.
For a "pro touring" type car like you're describing, it'd be ultra-hard to beat the mentioned LS motor/trans combo. Probably a fairly common and relatively easy swap, though and it looks as if you're wanting something unique.
That's awesome that it's a family car and you're building it how you want it. ChevyCrew is dead nuts right on the fix it yourself factor of a carbed, HEI older motor though. Not much to go wrong with one of those.
